FAQ - 💬❓ What is AmBisome used for?👉 This medication is used to treat a variety of serious fungal infections. It is often used in patients who cannot tolerate or who do not respond to the regular amphotericin treatment. It works by stopping the growth of fungi. ❓ What type of drug is AmBisome?👉 AmBisome is an antifungal medication that fights infections caused by fungus. AmBisome is used to treat serious, life-threatening fungal infections including leishmaniasis, or a certain form of meningitis in people infected with HIV (human immunodeficiency virus). ❓ Is AmBisome an antibiotic?👉 Amphotericin B is a macrocyclic, polyene, antifungal antibiotic produced from a strain of Streptomyces nodosus. ❓ How do you administer AmBisome?👉 AmBisome should be administered by intravenous infusion over a 30 - 60 minute period. For doses greater than 5mg/kg/day, intravenous infusion over a 2 hour period is recommended (see section 4.4). The recommended concentration for intravenous infusion is 0.20 mg/ml to 2.00 mg/ml amphotericin B as AmBisome. ❓ What is the major side effect of amphotericin B?👉 The principal acute toxicity of AmB deoxycholate includes nausea, vomiting, rigors, fever, hypertension or hypotension, and hypoxia. Its principal chronic adverse effect is nephrotoxicity. AmB probably produces renal injury by a variety of mechanisms. ❓ Why is amphotericin B so toxic to humans?👉 Mechanism of toxicity Amphotericin B molecules can form pores in the host membrane as well as the fungal membrane. This impairment in membrane barrier function can have lethal effects. Ergosterol, the fungal sterol, is more sensitive to amphotericin B than cholesterol, the common mammalian sterol. ❓ What is the most serious side effect of amphotericin?👉 The principal acute toxicity of AmB deoxycholate includes nausea, vomiting, rigors, fever, hypertension or hypotension, and hypoxia. Its principal chronic adverse effect is nephrotoxicity. ❓ What drugs should not be taken with amphotericin?👉 Some products that may interact with this drug include: anti-cancer drugs (such as mechlorethamine, nitrogen mustard), azole antifungals (such as ketoconazole, itraconazole), cidofovir, digoxin, flucytosine, medications that affect the kidneys (including pentamidine, tacrolimus, aminoglycosides such as gentamicin), ... ❓ What is the major side effects of amphotericin B?👉 With intravenous injection
❓ Why does amphotericin B affect the kidneys?👉 Amphotericin B binds to sterols in cell membranes, thereby creating pores that compromise membrane integrity and increase membrane permeability. It binds not only to ergosterol in fungal cell walls but also to cholesterol in human cell membranes; this is what accounts for its nephrotoxicity. ❓ What is the major downside to long term treatment with amphotericin B?👉 In cancer patients with persistent fever and neutropenia, amphotericin B is administered for the early treatment and prevention of the clinically occult invasive fungal infections. The major drawback of the conventional amphotericin B is its nephrotoxicity. ❓ What is AmBisome used for?👉 AmBisome is an antifungal antibiotic. The active ingredient in AmBisome is amphotericin B. AmBisome is given as an infusion into a vein (a drip) in hospital by a doctor or nurse. AmBisome is used to treat serious infections caused by fungi: Fungal infections of one or more deep organs of the body. ❓ What is the structure of AmBisome?👉 AmBisome consists of these unilamellar bilayer liposomes with amphotericin B intercalated within the membrane. Due to the nature and quantity of amphophilic substances used, and the lipophilic moiety in the amphotericin B molecule, the drug is an integral part of the overall structure of the AmBisome liposomes. ❓ What is AmBisome® (amphotericin B)?👉 Mechanism of action of AmBisome® (amphotericin B) liposome for injection. AmBisome is a true single-bilayer liposomal drug-delivery system1. Cross-section view of liposome. Liposomes are closed, spherical vesicles created by mixing proportions of amphophilic substances (such as phospholipids and cholesterol) ❓ What are the possible side effects of AmBisome?👉 Kidney damage is a frequently reported side effect, and can be severe and/or irreversible. Less kidney toxicity has been reported with liposomal formulations (such as AmBisome) and it has become preferred in patients with preexisting renal injury. |
